Once you connect 2 teleporters, you need to place any trigger mechanism that will make your teleport work correctly. There is no difference in what trigger to choose, but it is recommended to opt for a Pressure Plate. It is perfectly suitable for teleporting.

You only need to interact with the trigger, and all mobs in the 3×3 area will be teleported to the other side of the teleport. Also, keep in mind that a 3×3 area also works in height. So, if you want to teleport only ground units, place the required teleporter 2 layers underground.  

And the last thing you should know is that teleporting always works 2-way. Mobs, enemies, and players from both sides will be teleported as soon as the trigger is activated. Therefore, it would be best to place the teleporter remotely to avoid accident teleporting.
