Unfortunately, it is not possible to save your game manually without quitting Animal Crossing: New Horizons as well, so you should be using manual saves only when you are also going to quit the game. Also, do note that it is not possible to save the game and quit if you have friends visiting your island, so you will have to wait for them to leave before you can do so.
